Role for polo-like kinase 4 in mediation of cytokinesis

A cell-line study examines PLK4 localization and cytokinesis through antibody, genetic and inhibitor assays. The cleavage mechanism and cellular inhibitor specificity remain uncertain. Figure captions, sample panels and starting-cell counts conflict, while some replicate and animal-method details are missing. Nolan contributed reagents and analytic tools. Funding, intellectual property and commercial disclosures are specifically attributed.

What we checked

All 137 supplied XML units and 49 references were read in four untruncated returns. Six meaningful main JPG figures and all eleven static appendix pages were inspected, including supplementary figures S1–S5 and six movie descriptions with static previews. Root checked the native eleven-author list, actual unit headers, appendix page count and frozen predecessor bytes. Six offered MOV files were received but remain entirely unviewed, each before frame 0/time 0. Their reported cell counts are caption testimony, not observed dynamics. The original main PDF layout, historical versions, fine glyphs and raw experimental records remain incomplete.

Complete supplied main and static appendix read; movies unviewed

How we check sources

Not checked yet

  • Six received movies remain wholly unviewed. Static previews and caption counts do not establish continuous playback or independent cell counting. Main original PDF layout and historical corrections remain unreviewed.
  • Exact unprinted plot values, band intensities and some tiny domain/scale labels are unknown. Fig5E caption says OVCAR3 but its image says KOC7C; initial one-million versus 200,000-cell protocols and supplementary panel/caption statistics conflict.
  • Cleavage sites and protease are inferred. A cited cellular-selectivity criticism remains unread; biochemical IC50 arguments do not independently prove cellular specificity. Sample/replicate/error-bar definitions are incomplete and repeated lines are dependent.
  • Xenograft species, animal counts, allocation, methods, ethics and tissue custody are absent from the supplied scope. Raw blots, sequences, assays and counted fields were not acquired. No clinical validation or independent replication is established.

Source limitations

  • Nolan is credited with new reagents/analytic tools. Mak equity and IP transfer, Slamon industry interests and CIRM/NCI-core/foundation support are retained. Funding and affiliations do not establish a UAP programme.
