Functional comparison of PBMCs isolated by Cell Preparation Tubes (CPT) vs. Lymphoprep Tubes

A paired laboratory comparison uses three donors, three operators and eighteen processing aliquots to compare CPT and Lymphoprep methods. Differences in collection and centrifugation form part of the method packages. Nonsignificant results do not establish equivalence. Tables contain endpoint exceptions to broad conclusions, and figure captions and antibody lists conflict. Nolan contributed design, analysis and writing; commercial affiliations and funding are specifically attributed.

What we checked

All 115 supplied XML units and 15 references were read, including 13 separately recovered abbreviation labels. All 13 current publisher PDF layouts and four meaningful main figures were inspected. Main Table 1 includes 48 native rows and 674 cell fields across PDF pages 4–6. Both unique offered supplements were read: a one-page Figure S1 PDF and all 119 workbook cells, comprising 58 nonempty and 61 blank fields. Root independently matched workbook values, types and stored numeric lexemes; no formulas were present or evaluated. Native spreadsheet visual formatting, missing S1 axis exponents, tiny flow ticks, unprinted values and historical versions remain unresolved. Duplicate descriptions and four unviewed GIF thumbnails receive no additional credit.

How we check sources

Not checked yet

  • Native Excel visual formatting was not inspected. Figure S1 count exponents are missing or unreadable; they were not inferred from main figures. Tiny ticks and exact unprinted points remain unknown.
  • Three donors at one time point constrain generalization; eighteen aliquots are not eighteen independent people. The additional supplement set’s donor overlap is unspecified. No equivalence margin, power analysis, confidence interval or independent replication establishes clinical equivalence.
  • Pre-freeze method yield has ANOVA p=.0256 versus Wilcoxon p=.0977. Some method/operator endpoint contributions qualify aggregate donor-dominance claims. NKT table statistics are absent; Fig3 G/H captions and antibody/figure marker lists conflict.
  • Raw data available on request were not obtained; no private volunteer records, consent forms, code execution or new clinical advice were collected. Historical revisions, corrections and cited originals remain unreviewed.

Source limitations

  • Nolan is credited with design, analysis and manuscript contribution; other authors generated the data. WCCT/ARK/Primity affiliations, NIH/WCCT funding and the no-conflict declaration are recorded together. No government approach or UAP programme is established.
