Ultra-high throughput single-cell analysis of proteins and RNAs by split-pool synthesis
The authors demonstrate split-pool barcoding for protein and targeted RNA measurements in cell lines, human blood-cell material and mouse cells. Large theoretical barcode capacity is distinct from measured event recovery. Uneven RNA detection, cell losses, exclusions, doublets and source discrepancies limit interpretation; no clinical or UAP validation follows.

How we check sources
Not checked yet
- Twelve information-page visuals, main Figure9 fine alignment glyphs, original main PDF, raw data, code and current patent status remain unreviewed. The received workbook was unopened and unread. Correction-listed figure changes were not described precisely, so version concordance remains unknown.
- Filtered detected cells and same-preparation aliquots are not independent donor cohorts or all barcoded cells. Failed experiments and limited replication leave an unknown attempted-experiment denominator. RNA absence can reflect assay loss or detection limitations.
- Marker labels, cell counts, target numbers, repository identifiers and reporting-form human/flow categories conflict within the supplied sources. Commercial financial interests and patents are disclosed. Nolan has substantial originating, supervision, design and writing roles; these do not validate unrelated claims.
