Multi-omic single-cell snapshots reveal multiple independent trajectories to drug tolerance in a melanoma cell line
The authors infer different routes to early drug tolerance within one cultured melanoma cell line and test selected drug combinations. The inferred barrier was not measured. Negative inhibitor assays, dependence among cell snapshots and possible normal-tissue toxicity qualify the results; clinical efficacy was not established.

- One patient-derived cell line and674 dependent measured cells do not constitute a clinical cohort or independent donor trials. Trajectories, marker networks and the barrier are model inferences. Inhibitors without BRAFi had negative short-term growth/toxicity results under the reported assay.
- Unexplained timing and trace-count differences, a module1/2 caption mismatch, reporting checkbox/software discrepancies and unresolved contributor initials remain. Shared conceptual advice does not make Nolan the sole experimental lead or validate unrelated UAP claims.
