Multimodal and spatially resolved profiling identifies distinct patterns of T cell infiltration in nodal B cell lymphoma entities

This lymphoma study maps T-cell composition across 101 lymph-node samples and imaging specimens from 19 patients. Cells, cores and repeated assays remain dependent. External cohorts associate signatures with survival, but thresholds are data-derived and endpoint wording conflicts remain. Spatial proximity and inferred trajectories do not establish mechanism or clinical accuracy. Nolan contributes resources, supervision and funding acquisition; commercial roles and research support do not establish a classified UAP approach.

What we checked

A specialist read all 90,029 supplied XML characters in 11 contiguous untruncated returns, including seven main and ten extended figure captions and 69 references. Root reviewed the entire final analytical packet, not the complete primary text or figure pixels. Native figures/PDF, supplements, statistical source data, patient-specific metadata/charts, datasets, code, the patent, reviewer reports, external cohort originals and current correction history remain unreviewed in this lane.

Complete specialist supplied XML text; native apparatus unreviewed

How we check sources

Not checked yet

  • All 17 figure captions were read; figure pixels and the native journal PDF/layout were not viewed.
  • Supplementary Figures 1–2, Tables 1–6, the Reporting Summary and 17 statistical source-data files were not acquired.
  • Patient-specific supplementary metadata and private clinical charts were not acquired.
  • External cohort originals and raw data were not read in this lane. Survival-curve participant counts, hazard ratios and confidence intervals are not established by the supplied text.
  • GEO GSE252608/GSE252455, BioStudies S-BIAD565 and figshare 10.6084/m9.figshare.24915633 are availability statements; datasets were not downloaded or audited.
  • The interactive imaging browser was not opened. Linked code and the patent original were not retrieved or run.
  • All 69 reference entries were read; that does not establish reading of their original papers. Earlier independently accepted source scopes remain.
  • Named reviewers are acknowledged, but their reports were not read. The complete publisher version and correction history were not audited.
  • Complete supplied text does not complete the illustrated article, all Nolan material, original recordings or the whole project.
  • 101nodes=28DLBCL/30FL/15MZL/15MCL/13reactive controls; reactive nodes are not an unselected healthy population. CITE51/pairedflow13/independentflow50/TCR17 and24sample IKZF3 validation are not additive unique participants; exact overlaps retained.
  • Imaging35cores from19patients, at most two cores per patient; millions of cells are nested. Some core-level proportions lack an explicit repeated-patient model. Five-and-a-half-million processed versus5,690,284 initial cells and50antibody/52marker wording remain qualified.
  • Cell-marker boosting uses cell-level tenfold folds without explicit donor blocking. Separate sample-level nested leave-one-out entity classification reports74.5% balanced accuracy; authors do not claim diagnostic accuracy or external entity-classifier clinical validation.
  • Independent retrospective DLBCL and prospective-GALLIUM-derived FL bulk data show observational survival associations. Data-derived maxstat cutoffs and literal maximized P value wording remain; locked threshold, FL survival n, hazard ratios/confidence intervals unestablished here. Genetic-analysis190/137counts are not every survival denominator.
  • FL regulatory-cell survivalP0.17 is null and helper-cellP0.05 borderline; corrected genetic comparisons allP>0.05 despite displayed uncorrected results. ABC/GCB contrast is inconsistent across cohorts. Nulls do not prove equivalence.
  • ExtendedData7 overall-survival title versus body/method progression-free endpoint, and d/e helper/regulatory panel references conflict. Native curves unviewed; do not choose a preferred endpoint or silently repair references.
  • Pseudotime, inferred transcription factors, shared clonotypes and spatial proximity are associations, not lineage tracking, gene perturbation, active signalling or tested response to proposed therapy. Prior CAR-T/bispecific/transplant recipients excluded.
  • No sample-size determination/randomization/blinding. Statistics no-data-excluded statement differs from earlier viability, treatment, contamination and cell/image-quality filtering; preanalytic/downstream scopes unreconciled. Treatment/age confounding retained.
  • Nolan Resources/Supervision/Funding acquisition roles do not assign every grant to him or establish personal assay work. Correspondents/joint-supervisors Huber/Dietrich. Exact named BMBF/DFG/Heidelberg/EMBL/other recipients and Roche GALLIUM sponsorship preserved; original contracts unreviewed.
  • Nolan declares Akoya cofounder/stockholder and US9909167 inventorship; Schürch EnableMedicine advisory/options/funding. Article-time disclosures are not current registry audit, proven bias, clinical treatment or classified UAP programme evidence.
