Spatially Segregated Macrophage Populations Predict Distinct Outcomes in Colon Cancer

This observational human-tissue study associates distinct macrophage populations with different colon-cancer outcomes: IL4I1 with favorable survival in two reported cohorts and SPP1 with unfavorable survival in one. Nested cells and tissue regions are not independent patients; no adjusted or prospectively validated clinical predictor is established. A reused breast-cancer treatment-response measure is T-cell receptor expansion, a surrogate. Nolan is a Resources coauthor. Public cancer-research grants name other recipients and establish no classified approach or UAP programme. Figures and supplements remain unreviewed.

What we checked

Specialist read all 102,777 supplied XML text characters in 11 contiguous untruncated returns, including seven figure captions, 12 supplementary descriptions, declarations and 63 reference entries. Root reviewed the complete frozen analytical report and evidence; root did not read the full original source. Figure pixels, supplementary files, external data/code, peer review, current correction history and cited originals remain unreviewed.

Complete specialist supplied XML text; native graphics unreviewed

How we check sources

Not checked yet

  • Seven native graphics
  • SupplementaryTablesS1-S3 contents
  • SupplementaryFiguresS1-S9 pixels/full contents
  • External viewer high-resolution image data
  • External patient-level datasets/tabulations
  • External scripts and reanalysis
  • Peer-review reports
  • External version/correction history
  • Original references
  • Exact survival-cohort demographics/Ns/effect sizes not recoverable from supplied text alone
  • Complete supplied XML text is specialist consumption, not complete root reading, native figure review or the whole illustrated article. Bibliographic entries do not establish reading 63 cited originals.
  • The discovery reuses four public studies: 18,698 cells, 128 samples and 92 patients. Multiple modality and tissue-region subsets overlap and must not be added into a new patient count. Tissue-region random intercepts are described; patient-level nesting across regions is not explicitly modeled.
  • IL4I1 survival association is reported across TA118 and TA379; SPP1 is assessed only in TA118. Higher-cell-count core selection and top/bottom 40% Kaplan-Meier comparisons are retained. No specified Cox covariate adjustment, prespecified cutoff, holdout, prospective validation, blinding or sample-size power calculation establishes a clinical predictor.
  • The reused pembrolizumab response label refers to T-cell receptor expansion, not demonstrated tumor regression or survival. Mechanistic and therapeutic interpretations are not intervention-based causal proof.
  • Nolan is a Resources coauthor, not a corresponding author. Funding names West/van de Rijn, Hickey and Lu; no Nolan-specific award, classified government approach, UAP subject or anomalous material is established. Article-specific disclosures do not establish universal absence of interests.
  • Native 14-versus-13 neighborhood counts, 0.5-versus-1 square millimeter regions, CD63-versus-CD163 labeling, figure-panel references and 98% total proportions remain unresolved. Publication March27,2024 differs from print August2 and discovery-index August1; current substantive version history is unchecked.
