Sex Differences in the Blood Transcriptome Identify Robust Changes in Immune Cell Proportions with Aging and Influenza Infection

The authors derive a144-gene sex-expression signature from public cohorts and explore age, inferred immune composition and influenza response. Cohort reuse and repeated observations limit independent sample counts. Associations do not establish hormone mechanisms or clinical prediction. A retraction linked inside a cited reference concerns that older paper’s epistasis interpretation; it is not a retraction of this study.

What we checked

Read90,553 XML characters in12 complete returns,80 references,342 physical table cells with17 blanks and seven figure-object captions. Viewed the graphical abstract and six main figures, plus all eight S1 supplement text-and-pixel pages. Inspected main PDF pages0,11,17. Nolan generated a cohort with coauthors and contributed to the manuscript. The supplied main declares no competing interests. The early mistaken retraction attribution is explicitly corrected.

Complete supplied main, tables, meaningful figures and eight supplemental pages

How we check sources

Not checked yet

  • The supplementary workbook, other main and duplicate-supplement layouts, dense fine figure glyphs, original cited-notice XML/PDF, raw repositories and code remain unread or unreceived. The missing Fisher-distribution symbol is a native source lacuna, not silently supplied. No patient records or scientific code were collected or executed.
  • Native discovery counts458 versus447, study/sample totals, cohort counts and AUC values conflict or use different units. Expression-derived sex labels overlap classification evidence. Age comparisons are cross-sectional; inferred composition is not direct cytometry. Influenza challenge samples repeat observations from29 selected people. A male response AUC interval extends above1 as printed; this is not calibrated clinical validation.
  • SupplementS6 captions conflict with nonsignificant male plots and the sign of a CD4 association. Vaccine-response comparisons are nonsignificant. The cited Hemani notice concerns epistasis false positives; reuse of its expression cohort neither automatically validates nor invalidates this analysis. No misconduct, main-paper retraction, clinical benefit or UAP programme is established. Funding and roles are attributed to their stated recipients.
